I: Receipt of client DNA
  • Clone DNA into a specified vector and transfect it into BL21 DE3 cells
  • Clone DNA into a specified vector and transfect it into HEK293 cells
II: Pilot 50ml production
  • Small scale expression screen
  • Optimisation of growth/expression parameters followed by pelleting down and collecting cells
  • Small scale expression screen
  • Scale-up in shaker flask culture
III: Purification of cells from initial pilot
  • Cell pellets lyzed using a sonicator
  • Supernatant will be collected and will undergo affinity purification using Nickel or another appropriate affinity tag
  • Harvesting of cell culture supernatant
  • Confluent supernatant harvested and concentrated by cross flow filtration
IV: QC assessment of protein purity
  • Analysis of purity by SDS-PAGE, WB, and anti-His WB
